Making Microscope Slides at Home: A Guide to Onion Epidermis and Pollen

When your microscope arrives, the biggest letdown isn't image quality; it's figuring out what to look at. More beginners than you'd imagine end up staring blankly at an empty slide. The truth is, your kitchen and balcony are an endless source of specimens, as long as you know how to prepare a slide.

The core supplies for slide preparation are just three things: a slide, a coverslip, and a drop of clean water. A temporary wet mount simply means placing your sample in that drop of water, lowering a coverslip over it, and letting the liquid spread into an even, thin layer. That's all there is to it, but each step has its finer points.

Your first specimen: onion epidermis. Peel a scale leaf from an onion and use tweezers to tear off a small piece of transparent membrane from the inner surface (the purple layer is easiest to peel and comes with its own color). Place a drop of water in the center of a slide, lay the membrane flat in it, and lower a coverslip on top. At 40X you'll see neatly arranged brick-like cells; at 100-400X you can make out cell walls, nuclei, and vacuoles. This is the first lab in biology classes worldwide because it's perfect: thin, transparent, clearly structured, and costs nothing.

Second: pollen mounts. Pick a flower from your balcony, grip the anther with tweezers, and gently shake it in the water drop on the slide. Pollen grains scatter into the water; add a coverslip and observe. Pollen shapes vary enormously between plants: lily pollen is oval with furrows, while sunflower pollen is a spiky sphere. The three-dimensional sculpturing on the pollen wall is highly photogenic at 400X, making it excellent material for microphotography.

Third: pond water (or pickle brine, or aquarium water). Place a drop directly on a slide and observe; a coverslip is optional (though it helps). That single drop is a tiny ecosystem: paramecia, rotifers, algae, and amoebas take turns making an appearance. The trick is to sample water with a story: the surface film, the base of aquatic plants, or anywhere rich in organic matter will have a high density of organisms.

Three general tips: when lowering the coverslip, set it down slowly at a 45-degree angle from one side to drive out air bubbles (bubbles are the black holes ruining beginners' images); if there's too much water, draw it off from the edge of the coverslip with a tissue, and if there's too little, add more from the edge; and always use less sample rather than more. A thin layer gives the clearest view, while a clumped sample has no focal plane.

For a more advanced approach, try staining your mounts: iodine for onion epidermis (nuclei turn dark brown and stand out instantly), methylene blue for cheek epithelial cells (a classroom classic). But master the temporary wet mount first, and you'll find that the joy of seeing clearly far outweighs the thrill of staining. A ticket to the microscopic world costs just one drop of water and one slide.
