Ten Common Microscope Mistakes Beginners Make

Every rule for using a microscope exists because someone learned it the hard way. Here are the ten most common beginner mistakes. See how many you recognize.

Mistake one: jumping straight to the highest magnification. The field of view at high power is tiny, so when you cannot find your target you start blaming the instrument. The right approach: always start at the lowest magnification, find your target, move it to the center, then work your way up.

Mistake two: driving the objective lens down with the coarse focus knob. Lowering the objective while looking through the eyepiece is how coverslips get crushed and objective lenses get scratched. The right approach: watch from the side while lowering the lens, and only look through the eyepiece as you raise it.

Mistake three: wiping lenses with tissue or your shirt. Wood fibers and dust from fabric are what kill lens coatings. The right approach: use an air blower and lens paper, and apply cleaning solution sparingly only when necessary.

Mistake four: viewing without a coverslip at high magnification. Objectives of 40x and above are designed for a 0.17 mm coverslip, so image quality drops sharply when you view a bare slide. The right approach: always use a coverslip when preparing a slide, especially at high magnification.

Mistake five: letting a dry objective pick up cedar oil. After using the oil immersion objective, you rotate the nosepiece and the neighboring 40x lens brushes against the oil. The right approach: clean the oil immersion objective immediately after use, and keep an eye on the adjacent lens when switching objectives.

Mistake six: opening the aperture all the way. The idea that brighter means clearer is a misunderstanding. With transparent specimens, too much light actually washes out the detail. The right approach: set the aperture to roughly 70 to 80 percent of the objective's NA for the best contrast and resolution.

Mistake seven: using maximum brightness for live specimens. Under strong light, paramecia stop moving within minutes. The right approach: use medium to low brightness for live observation, and shorten your viewing time when you need more light.

Mistake eight: leaving slide fluid on the stage. Salt solutions and stains corrode the stage surface and the condenser. The right approach: use only as much fluid as you need, and wipe up spills immediately.

Mistake nine: putting the microscope away carelessly. The objective is left at high magnification, the brightness is still at maximum, and the dust cover never goes on. The right approach: follow a shutdown routine. Lower the stage, rotate to the lowest magnification, turn the brightness down to zero, and put on the dust cover.

Mistake ten: carrying the microscope against your chest. One bump can shift the collimation and ruin the image. The right approach: support the base with one hand and hold the arm with the other, and set it down carefully.

Get all ten of these right and your microscope will hold its image quality and last at least five years longer. Don't ask how we know. Behind every one of these mistakes is a microscope that paid the price.
