How to Focus a Microscope Correctly: Low Power First, High Power Later, and Never Crush the Slide

The first lesson for a microscope beginner is not what to look at, but how to focus. Crushed coverslips, an objective lens jammed against the slide, a completely black field of view that makes you think the instrument is broken, all of these accidents trace back to the same root cause: focusing in the wrong order.

The standard focusing procedure has five steps. First, place the specimen in the center of the stage aperture and secure it with the stage clips. Second, confirm that the lowest-power objective is in position, usually 4x. Third, watch the gap between the objective lens and the slide from the side while you turn the coarse focus knob to bring the objective closer to the slide. Note that you lower it while watching, not blindly.

Fourth, move your eye to the eyepiece and slowly turn the coarse focus knob in the opposite direction so the objective gradually moves away from the slide until an image appears, even a blurry one. Fifth, use the fine focus knob to sharpen it. The heart of this procedure is a single rule: watch from the side while the lens goes down, and look through the eyepiece only while it comes up. That alone reduces slide-crushing accidents to nearly zero.

The rule for switching to a higher-power objective: when you move from 4x to 10x or 40x, the objective is longer and the working distance shorter, so in theory you should first confirm the specimen is centered in the field of view, then rotate the nosepiece directly. Modern microscopes are designed as parfocal, so after switching you only need a slight turn of the fine focus knob. Still, the first time you use your instrument, check from the side that the high-power objective will not touch the coverslip.

If the field of view is completely black, check these in order: whether the aperture diaphragm is open, whether the condenser is set too low, whether the objective is properly clicked into position, whether the sample is actually in the light path, and, at high magnification, whether the coverslip is thicker than the standard. Nine out of ten black screens come from these five items and have nothing to do with whether the microscope itself is broken.

If the image always seems just a little off no matter how you focus, check two things. One, the fine focus knob may have reached the end of its travel; some models have a limited range, so turn it back a few rotations and try again. Two, the eyepiece diopter may not be matched to your eyesight. This is not an issue with a monocular, but with binocular eyepieces you should close one eye and adjust the sharpness for the other eye first.

One last habit worth adopting: when you finish observing, rotate the nosepiece back to the lowest-power objective and lower the stage before putting the microscope away. The next person, or tomorrow's version of you, can pick it up and start right away, and a high-power objective left hanging out in the open is less likely to get knocked. A microscope is a precision instrument; treat it by the book and it will behave for a decade.
