Slides and Coverslips: The Two Accessories Beginners Most Often Get Wrong

The microscope image looks blurry, you spend ages checking the lens and find nothing wrong, and then it turns out the slide was the problem. This happens to beginners far too often. Slides and coverslips are a team: used correctly, they protect image quality; used incorrectly, they are the source of every problem.

First, keep the two roles straight. The slide (the thick piece, about 1 mm) carries the specimen, so it is the tray. The coverslip (extremely thin, a standard 0.17 mm) flattens the specimen and the liquid, so it is the lid. The working distance of high-power objectives (40x and above) is designed around a 0.17 mm coverslip. A coverslip that is off by 0.02 mm in thickness already produces a visible difference in image quality, and if you observe directly at 40x without a coverslip, the image will always fall short.

The three mistakes beginners make most often: first, putting the coverslip on upside down or substituting something thicker, such as a plastic sheet or clear tape. The thickness is wrong, and high-power imaging collapses. Second, continuing to use a cracked coverslip. The image distorts along the crack, and it can easily scratch the objective lens. Third, failing to drive out air bubbles. Those black rings in the image are bubbles, yet people study them for ages thinking they are cells.

The standard way to apply a coverslip: after placing your sample on the slide, hold the coverslip by its edges, not the center, and touch one edge to the drop of liquid at a 45-degree angle, then lower it slowly. This motion pushes the liquid from one side to the other and drives the air out. Lower it in a hurry and the bubbles stay in.

Cleaning and storage: rinse used slides with water, scrub them with a mild detergent, rinse again, then dry with a soft cloth or let them air dry. Stubborn stain residue, such as methylene blue or iodine solution, can be wiped off with alcohol. For storage, stand them upright in a slide box rather than stacking them, which causes rubbing. Those fine scratches on the surface of a slide are the handiwork of stacked storage.

Coverslips are consumables. If one breaks or gets scratched, replace it; a box costs only a few dozen yuan and is not worth rescuing. When buying, however, make sure the thickness is the standard 0.17 mm plus or minus 0.02 and that the product is intended as a coverslip. Some sellers mix slides and coverslips together and label the dimensions unclearly, so you only discover the problem once the package arrives.

One last advanced point: coverslips also come in a high-precision thickness version (0.17 mm plus or minus 0.005, marked No. 1.5H), intended for users doing high-power photography above 40x and oil immersion work. For ordinary observation, the standard product is enough. If you are starting to take microphotography seriously, upgrading your coverslips is the most noticeable improvement you can buy for a few dozen yuan.
