4x, 10x, 40x, 100x Objectives: The Distinct Role of Each Magnification
The lenses on a microscope's revolving nosepiece are not simply the same thing at different magnifications. They are four specialists, each with a clearly defined role. Understanding what each one is for is the dividing line between owning a microscope and actually knowing how to use one.
4x (scanning objective): the widest field of view and the longest working distance, built for reconnaissance. Use it to see the whole specimen, locate your target, or observe the overall structure of an insect. The image is bright with deep depth of field, making this the first stop after every slide is prepared.
10x (low-power objective): the workhorse of efficiency. The field of view is still generous, but the amount of detail doubles. This is the main magnification for scanning slides, examining plant tissue structure, and viewing larger microorganisms. In day-to-day use, it often sees the most action of any objective.
40x (high-power objective): the detail specialist. This is the primary battleground for animal cells, plant cells, and protozoa. Note that its working distance has already dropped below 1 mm, so a coverslip thicker than 0.17 mm will directly affect image quality.
100x (oil immersion objective): the one that pushes the limit. Use it for bacteria, chromosomes, and the fine details of organelles. It must be used with cedar oil (or synthetic immersion oil) between the objective and the coverslip. The refractive index of the oil is close to that of glass, allowing a high-numerical-aperture objective to truly take in light. Without oil, the image turns to mush; if you do not clean it after use, the objective is ruined.
Besides magnification, the objective barrel carries a string of numbers, and the most valuable of these is NA (numerical aperture). NA determines the limit of resolution: an NA of 0.65 can resolve details down to roughly 0.8 microns, while an NA of 1.25 reaches 0.4 microns. Among objectives of the same magnification, a higher NA means a higher price and a sharper image. When buying, this is a harder specification than brand name.
There are two more markings worth knowing. The color ring: different magnifications use different colors for quick identification. The marking "/0.17" indicates the objective is designed for a coverslip thickness of 0.17 mm. The symbol "∞" means an infinity-corrected system, which forms an image in combination with the tube lens and is the mainstream choice for mid-range and high-end models.
A mnemonic for use: 4x shows you where, 10x shows you what, 40x shows you structure, and 100x shows you detail. Do not skip steps, or you will lose your target. Follow the order, and every observation becomes an adventure that goes deeper layer by layer.
